cell from the adult mouse pancreas (.01%) that can show extensive proliferation under completely defined conditions\u00a0<em data-w-id=\"c4460e1b-bdbb-b419-df8c-05c88c876219\" data-wf-id=\"[&quot;c4460e1b-bdbb-b419-df8c-05c88c876219&quot;]\">in vitro<\/em>\u00a0(Seaberg et al, 2004). \u00a0Each single, adult pancreatic multipotent precursor (PMP) cell produces progeny from two separate organ lineages (and only those lineages): first, exocrine and endocrine pancreatic lineage cells (including B-like cells) and second, neurons and glial cells. \u00a0We suggest that\u00a0<em data-w-id=\"e34c5996-f27a-5371-5a27-e16d329b8fe9\" data-wf-id=\"[&quot;e34c5996-f27a-5371-5a27-e16d329b8fe9&quot;]\">in vivo<\/em>\u00a0PMPs only make pancreatic lineage cells, but in vitro the expression of a toolbox of transcription factors that are important in both pancreatic and neural development permits the PMPs to make neural cells as well. \u00a0We hypothesize that these cells may comprise a population of adult mammalian pancreatic stem cells, which might in the future be employed in treating type 1 diabetics.<br data-w-id=\"b6618b3c-5378-36bc-55dd-20a9fca24852\" as Notch\u00a0<em data-w-id=\"3b45f01f-0949-9967-1f5c-6bd9ae2a2899\" data-wf-id=\"[&quot;3b45f01f-0949-9967-1f5c-6bd9ae2a2899&quot;]\">in vivo\u00a0<\/em>and Wnt, as well as concentrations and media components. \u00a0This is particularly important if we are to maximize the number of cells available for treatment in mouse diabetic models (see 3rd aim below) and eventually in humans. \u00a0Thus, we will undertake small molecule screens for compounds that increase in the survival and expansion through symmetrical cell divisions of pancreatic stem cells and increase differentiation of insulin producing beta cell progeny of pancreatic stem cells. \u00a0Second, an apparent adult human pancreatic stem cell with properties similar to that of the adult mouse pancreatic stem cell will be characterized in detail. \u00a0Third, the\u00a0<em data-w-id=\"f8632281-0594-ef82-5f92-377c0ec97d88\" data-wf-id=\"[&quot;f8632281-0594-ef82-5f92-377c0ec97d88&quot;]\">in vitro<\/em>\u00a0PMP colonies
